High-mobility group protein B1 (HMGB1) ist ein multifunktionelles Protein mit verschiedenen Rollen in den unterschiedlichen Zellkompartimenten. Im Zellkern fungiert HMGB1 als DNA-Bindungsprotein für die Gentranskription, Genregulation und DNA-Reparatur.[1](https://ressourcen.biomol.com/de/biomol-blog/hmgb1-elisa-kits-von-arigo-biolaboratories?hs_amp=true#References) Im Cytosol induziert HMGB1 Autophagie indem es mit Beclin-1 interagiert, was zu dessen Dissoziation von Bcl-2 mit anschließender Induktion der Autophagie führt.[2](https://ressourcen.biomol.com/de/biomol-blog/hmgb1-elisa-kits-von-arigo-biolaboratories?hs_amp=true#References) Im extrazellulären Raum wird HMGB1 passiv von nekrotischen Zellen freigesetzt und wirkt als Chemokin und DAMP (*Damage-associated molecular pattern*), um Entzündungsreaktionen auszulösen.[3](https://ressourcen.biomol.com/de/biomol-blog/hmgb1-elisa-kits-von-arigo-biolaboratories?hs_amp=true#References) Außerdem wird HMGB1 von aktivierten dendritischen Zellen und Makrophagen sezerniert und fungiert als Cytokin. Es fungiert als Ligand für RAGE, TLR2 und TLR4 und aktiviert so die Signalwege und die folgenden Entzündungsreaktionen.[4](https://ressourcen.biomol.com/de/biomol-blog/hmgb1-elisa-kits-von-arigo-biolaboratories?hs_amp=true#References) Das aus apoptotischen Zellen freigesetzte HMGB1 wird durch die von ROS (reaktive Sauerstoffspezies) -abhängige Oxidation neutralisiert und fördert somit die Immuntoleranz. 

## HMGB1 bei Erkrankungen

HMGB1 hat sowohl die Aufmerksamkeit von Grundlagen- als auch von klinischen Forschern auf sich gezogen, da es mit verschiedenen Pathologien wie Krebs, Sepsis, Arteriosklerose, chronischer rheumatoider Arthritis und Nephritis in Zusammenhang steht.[5-7](https://ressourcen.biomol.com/de/biomol-blog/hmgb1-elisa-kits-von-arigo-biolaboratories?hs_amp=true#References) HMGB1 wird als Nekrosemarker beschrieben, und ein erhöhter HMGB1-Spiegel gilt as ein guter Indikator für eine wirksame Krebstherapie.
